Proper care of IAPS with ILASS robot fluidics

Daily Maintenance

Morning

  • Check the fixed tips on the liquid handling arm (LiHa) for any damage.

  • Make sure the system liquid container is full, and the inlet tubing is below the liquid. Keep the carboy opening covered to prevent contamination from entering the system liquid.

  • Make sure the system waste container is empty and the tubing does not have any kinks.

  • Perform a Liquid System Flush

    • Consult the Infinium ROMA (IAPS with ILASS) Product Documentation for more details on selecting the appropriate option in Illumina Lab Automation Software Solution (ILASS).

    • Observe diluter syringes and tubing to check for air bubbles.

    • Repeat additional system flush routines as needed to make sure all air bubbles are removed.

  • Check liquid system for leakage, such as droplets leaking from fixed tips, leaks in syringes, or stray drops on worktable.

  • If the system liquid carboy runs dry, conduct repeated system flush routines to prime the fluidics lines after refilling the carboy.

Between workflow steps

  • Perform a Liquid System Flush between workflow steps after instrument is idle for more than two hours.

End of Day

  • Check the fixed tips on the LiHa for any damage.

  • Perform the Diluted Bleach Tip Wash to minimize potential growth in fluidics lines and to clean the tips exposed to DNA.

    • Consult the Infinium EX Assay Product Documentation for details on selecting the appropriate option in ILASS.

    • Perform the Diluted Bleach Tip Wash only once per day and use a 1:10 bleach dilution (equivalent to 0.5-1% sodium hypochlorite final concentration).

    • After the bleach wash, wipe each fixed tip using 70% EtOH on a Kimwipe.

  • Clean various components of the instrument.

    • Refer to the Infinium Consumables and Equipment List under section "Robot Cleaning Reagents" for a list of recommended cleaning agents.

    • Remove carriers and racks from worktable and clean using an approved detergent, antiseptic solution, or 70% EtOH and rinse with water as needed. Do not use bleach or a laboratory washing machine.

    • Clean the worktable, the outside of the wash station, and the inner and outer surfaces of the front safety door with 70% EtOH. Do not use bleach.

    • Empty system liquid waste container and clean with detergent or antiseptic solution. Clean outside of waste tubing with an approved detergent, antiseptic solution, or 70% EtOH.

  • Refill the system liquid container if needed.

  • Check liquid system for leakage, such as droplets leaking from fixed tips, leaks in syringes, or stray drops on worktable.

Weekly Maintenance

  • Check and finger tighten all 16 screws for the tubing going into and out of each of the eight 3-way syringe valves.

  • Check the plunger lock screws and syringe screws and tighten manually.

  • After checking the syringes, flush the liquid system using a recommended cleaning agent, followed by deionized water, and finally with system liquid.

  • Empty and clean the system liquid container and system liquid waste container with 70% EtOH and refill.

  • Clean all other liquid containers to prevent deposition of crystals and growth of microorganisms.

Water Circulator for Te-Flow

  • Check for proper fluid levels in the water circulator which controls the temperature of the Te-Flow.

  • Before each use, open the lid and make sure that water is above the heating coils. If the fluid level is low, replenish as necessary with distilled water prepared with anti-microbial additive. It is not necessary to drain the existing fluid.

  • Confirm that the lid is securely in place to minimize evaporation.

  • Refer to the Infinium EX Assay Documentation for details on setting up the water circulator and Te-Flow.
