The Illumina 16S Metagenomics Sequencing Library Preparation protocol is a two-step PCR workflow that amplifies the v3 and v4 regions of the 16S gene in a single amplicon for metabarcoding studies.

Compatible index primer kits are listed on the Nextera XT product page, below, under the Index Adapters heading. (Note: the Illumina 16S protocol does not use the Nextera XT library preparation kit, this link is only to reference compatible index primer kits.)
