The Illumina 16S Metagenomics Sequencing Library Preparation protocol is a two-step PCR workflow that amplifies the v3 and v4 regions of the 16S gene in a single amplicon for metabarcoding studies.
The protocol is available on the page here, along with additional supporting documentation and links to public data in BaseSpace.
Compatible index primer kits are listed on the Nextera XT product page, below, under the Index Adapters heading. (Note: the Illumina 16S protocol does not use the Nextera XT library preparation kit, this link is only to reference compatible index primer kits.)
