Input recommendations and considerations for COVIDSeq (RUO) library preparation

What input sources are supported for COVIDSeq library preparation (RUO)?

The Illumina COVIDSeq Test (RUO) supports samples derived from nasopharyngeal (NP), oropharyngeal (OP), nasal swabs, and wastewater.

How should samples be stored and shipped (if applicable) prior to library preparation?

Store samples according to the instructions from the manufacturer. Exceeding the storage times can negatively impact test results.

Transport samples according to the governing regulations for the transport of etiologic agents applicable to your region.

Should RNA be DNase treated?

Illumina does not have specific recommendations for this.

What mass of RNA should be used as input?

Illumina does not have guidance for this. Follow input recommendations for extraction process and volume recommendations for extracted RNA used in subsequent protocol steps.

Can cDNA be used as input?

Illumina has not validated this workflow with cDNA input.

See the Illumina COVIDSeq RUO Kits Reference Guide for additional information.
