How many samples can be prepared with the Illumina Exome 2.5 panel?
The Illumina Exome 2.5 Panel is used with the Illumina DNA Prep with Exome 2.5 Enrichment and Illumina FFPE DNA Prep with Exome 2.5 Enrichment library preparation kits.
Illumina DNA Prep with Exome 2.5 Enrichment
The Twist Bioscience for Illumina Exome 2.5 panel oligos tube contain enough material for 96 samples, including overfill, for automation. If preparing samples manually, there will be more than enough panel to cover additional hybridization reactions if desired. In the manual workflow, 96 samples requires 8 hybridization reactions at 4 µl each = 32 µl panel needed for 96 samples. The guaranteed fill volume is 68 µl.
Note that this library preparation kit only supports 12-plex enrichments, with each enrichment reaction requiring 4 µl of probe panel. One tube of the Illumina Exome 2.5 panel is provided per order of Illumina DNA Prep with Exome 2.5 Enrichment, (S) Tagmentation Set B (96 Samples, 12-plex) (catalog 20077595), or per order of Illumina DNA Prep with Exome 2.5 Enrichment, (S) Tagmentation Set D (96 Samples, 12-plex) (catalog 20077596). Index adapters plus UMI anchors are also included.
Illumina FFPE DNA Prep with Exome 2.5 Enrichment
For samples prepared with automation, 4 tubes of probes are required when preparing 192 samples to for sufficient overfill. Library preparation and enrichment reagents for 192 samples for automation can be purchased by ordering: Illumina Cell-Free and FFPE DNA Prep with Enrichment, Ligation (192 Samples, 4-plex), Cloud Analysis (catalog 20104103) or Illumina Cell-Free and FFPE DNA Prep with Enrichment, Ligation (192 Samples, 4-plex), On-premises (catalog 20104104). Index adapters plus UMI anchors and probe panel sold separately.
For samples prepared manually, each Illumina Exome 2.5 panel tube contains a volume of 68 µl, which is guaranteed to support 20 hybridization reactions. Each hybridization reaction will contain 4 samples (4-plex) and require 3 µl of probes. Therefore, 1 tube of probes can support 80 samples by the manual workflow.
