Troubleshooting NextSeq 500/550 run stopping with "Local Run Manager Error: Bad Request" error
The following message can appear during NextSeq 500/550 pre-checks and stop the run right when it begins:
"'Local Run Manager Error - Unable to setup impersonation with credential for \sbsuser' Originally caused by: bad request"
This message is typically related to the sbsuser Windows password being expired and requiring updating or the sbsuser password having been recently changed but the credentials not updated in Local Run Manager.
Note: Since the error appears during pre-check and stops the run while being initiated, the RFID for reagents are usually not recorded. Users are often able to set up the run again with the same consumables.
Troubleshooting steps:
Power cycle the instrument.
From the instrument home screen, select Manage Instrument.
Select System Shut down Options.
Select Shut Down (this will shut down software and turn off instrument power).
Wait two minutes before turning on the instrument from the button in front.
If prompted at sign in, update the credentials for sbsuser.
In the event the sbsuser account credentials require updating, it is likely the sbsadmin account needs updating as well.
To check if the credentials are expired and require updating:
Select Start then type
Run
to locate and open the Run app.Type cmd.exe followed by the Enter key. Open the Command Prompt window.
Type
net user sbsadmin
into the Command Prompt window followed by the Enter key.The output will detail the date the credentials were last changed and when they will expire.
If the admin credentials require updating:
Select the Windows Start Menu, then the User Icon.
Select Log Out or Switch Users.
Sign into the sbsadmin account then update the credentials.
Once complete, follow the same steps to log out of sbsadmin and back into sbsuser.
Update the new sbsuser account credentials in Local Run Manager.
For further information on updating the sbsuser account credentials in Local Run Manager, search for Knowledge Base article Updating Windows 10 credentials in NCS v4.0.1.+, MNCS v2+, MCS v4+.
Once the credentials are updated, confirm the NextSeq Control Software is able to initialize without error before attempting to set up the run again with the same set of consumables.
If consumables have been marked as used, a new set of reagents will need to be prepared. Contact Illumina Technical Support if there are any questions or concerns about doing so.
If the error is persisting after applying the above guidance or issues are encountered while working though the guidance, contact Illumina Technical Support for additional assistance.
If required as an interim solution, the run can be set up in Manual Mode as outlined in the NextSeq 500 and 550 Product Documentation.
