Background
The MiSeq i100 Series uses pressure assisted fluidics to move reagents through the cartridge and flow cell. This requires that the wet cartridge is pressurized to a specific pressure range, and must be maintained throughout the sequencing run.
If there are issues with the reagent seals on the wet cartridge, this can cause issues with the Pre-Run Checks or during the instrument sequencing runs. If this occurs during a sequencing run, the Control Software presents the following error:
The pressure charging failed due to air leakage detected. Contact Illumina Technical Support.
Troubleshooting Actions
Select Eject Cartridge, then remove the dry and wet sequencing cartridges from the instrument.
Power cycle the instrument to ensure all board communications are reset.
Run the instrument Mechanical and Fluidics System Health Checks using the Test Cartridges provided with the instrument.
Once the System Health Checks are complete, select Clear Checks and then proceed to the Control Software home screen.
Use the Export Logs - Small option to bundle the System Health Check results.
If applicable, select the checkbox next to any affected Sequencing Run(s) from the list to include the run Logs folder.
If this issue is observed, see the following Troubleshooting Actions and contact .
