What is ATAC-Seq? The assay for transposase-accessible chromatin with sequencing (ATAC-Seq) is a library preparation method for determining chromatin accessibility across the genome. Read more about ATAC-Seq here.
What reagents are available from Illumina? Illumina does not offer a commercial ATAC-Seq kit nor library preparation protocol. Custom ATAC-Seq protocols can be found in the literature and are not supported by Illumina.
Some custom ATAC-Seq library preparation protocols use Tagment DNA Enzyme (TDE1) and Tagment DNA Buffer (TD) buffer from the discontinued Nextera DNA Kit. TDE1 and TD Buffer can still be purchased as part of the Illumina Tagment DNA Enzyme and Buffer Small and Large Kit. The following table lists the kit contents:
Catalog Number
Contents
20034197 (Small Kit)
Tagment DNA Enzyme, 0.17 ml (1 tube)
Tagment DNA Buffer, 1.24 ml (1 tube
20034198 (Large Kit)
Tagment DNA Enzyme, 0.65 ml (1 tube)
Tagment DNA Buffer, 1.24 ml (2 tubes)
Use of these reagents in custom ATAC-Seq protocols will need to be validated by the user, and Illumina does not guarantee success.
Illumina provides volumes for these reagents instead of reaction numbers, because different ATAC-Seq custom protocols may utilize different reagent volumes per sample. TDE1 and TD Buffer are offered as a standalone bundle, and individual subcomponents cannot be purchased separately.
Other reagents used in custom ATAC-Seq protocols are purchased from third-party vendors. Illumina cannot provide recommendations for third-party reagent purchases.
