# Which lanes should be selected during sample sheet creation for a 1.5B run on the NovaSeq X/X Plus

The NovaSeq X/X Plus 1.5B flow cell uses a 2 lane configuration. When the reagents are loaded onto the instrument, the lyo tube will physically sit in alignment with lanes 4 & 5 on the flow cell.

Q: When creating a sample sheet during run setup for a NovaSeq X/X Plus 1.5B run, should the selected lanes be 1 & 2 or 4 & 5?

A: Lanes 1 & 2 should be selected during sample sheet creation for 1.5B runs.
