Washes on the MiniSeq

**The MiniSeq has three types of washes, listed below.

Automatic Post-Run Wash (60 minutes)**

  • When a sequencing run is complete, the software initiates an automatic post-run wash using the wash solution and NaOCl provided in the sequencing reagent cartridge. When the wash is complete, the Home button becomes active. Sequencing results remain visible on the screen during the wash.

  • After the wash, the sippers remain in the down position to prevent air from entering the system. Leave the cartridge in place until the next run.

Quick wash (20 minutes)

  • Required every 7 days the instrument is idle.

  • Requires 0.05% Tween 20 wash solution.

  • Load 40 ml wash solution into the wash cartridge in position 40 (indicated as B in image below).

  • From the Home screen, select Perform Wash, and then select Quick Wash.

Manual post-run wash (90 minutes)

  • Performed when a run ends prematurely, or when a flow cell (FC) saved for rehyb.

  • Load 1.3 ml of 0.12% NaOCl into position 31 (indicated as A in the image below).

  • Load 80 ml 0.05% Tween 20 wash solution into position 40 (indicated as B in image below).

  • From the Home screen, select Perform Wash, and then select Manual Post-Run Wash.

Steps for loading the wash flow cell and wash cartridge:

  1. Load the wash flow cell. Close the flow cell clamp and flow cell door.

  • Note: Alternatively, load a used flow cell.

  1. Remove the used reagent cartridge from the previous run, if present.

  2. Load the prepared wash cartridge.

  3. Remove the waste bottle and discard the contents in accordance with applicable disposal standards.

  4. Slide the empty waste bottle into the compartment until it stops.

  5. Close the reagent compartment door.

Wash Tray Care

  • Clean with deionized water and invert the wash cartridge when not in use to prevent microbial growth/contamination.

  • Note: The lid for the wash tray (well 40) can bend sippers if not placed correctly on top of the tray.
