Can NextSeq v2.5 flow cell be used with v2 Reagent Cartridge and Buffer Cartridge?
Yes, v2.5 kits include a v2.5 flow cell and v2 reagent and buffer cartridges.
In addition to starting intensities, are other run metrics expected to change?
Run specifications for the NextSeq v2.5 reagent kits, such as overall Q30 and output, are the same as for the v2 kits.
Can the NextSeq v2.5 reagent kit be used on the NextSeq 550Dx instrument?
Due to software incompatibility, NextSeq v2.5 reagent kits are for use only on the NextSeq 500/550 Research Use Only instrument at this time
Is there a visible change to the flow cell?
The flow cell part number and packaging labels are different; however there are no visible differences in the flow cell.
Can the v2 kit still be ordered?
NextSeq 500/550 v2 kits have been discontinued effective December 31, 2018; shipments continued through February 28, 2019. Illumina Advantage (TG) reagents versions can still be ordered.
Can the flow cell ID be used to distinguish between flow cell types?
No.
Are there any changes to loading?
Recommendation has changed for mid-output (MO) kits. NextSeq System Denature and Dilute Libraries Guide indicates 1.5 pM rather than 1.8 pM.
Are custom recipes for v2 kits usable with v2.5 kits?
No, custom recipes must be updated for v2.5 kits. NextSeq Control Software (NCS) v2.2 will not check if the recipe is correct (for v2 or v2.5) and will allow the run to continue to completion.
