Pre Run Check Troubleshooting on the MiniSeq
If the MiniSeq pre-ruck checks do not pass, follow the guidance below for the check or checks that did not pass.
General/reagent checks
Doors closed.
Make sure doors closed.
Power cycle* if issue persists.
Consumables loaded.
Ensure consumables are properly loaded.
Power cycle* if issue persists
Required Software.
Verify that the proper software/module is installed.
Instrument and network disk space checks - the control software will provide a warning if the free storage is below 100Gb.
Clear disk space from the D:\ drive.
Clear disk space from the network output location, if needed.
Network connection.
Confirms network status and physical network connection.
Check read/write permissions, availability of network location, and Ethernet plug.
Temperature
Temperature ramp: checks function of flow cell heater plate.
Power cycle* and retry the pre-run checks.
Temperature sensors**.
Power cycle* and retry the pre-run checks.
Fans**
Power cycle* and retry the pre-run checks.
Imaging System
Imaging limits**: moves stage to sensor limits.
Power cycle* and retry the pre-run checks.
Z step-and-settle**: checks the ability of z-stage to move and stop moving in a set time.
Power cycle* and retry the pre-run checks.
Bit Error rate: checks camera circuits.
Power cycle* and retry the pre-run checks.
Flow cell registration**: checks if the flow cell is in the correct imaging position.
Clean and re-seat flow cell and check gaskets.
Power cycle* and retry the pre-run checks. If the checks do not pass, try a new flow cell.
Reagent Delivery
Valve response**: tests the accuracy of valve and pump syringe rate movements.
Power cycle* and retry the pre-run checks.
Pump: moves pump syringe to min and max positions.
Power cycle* and retry the pre-run checks.
Note: Pre-run checks will not check flow rates, so always visually inspect the flow cell gaskets prior to loading.
