# Xp reagents components and stability on the NovaSeq 6000

NovaSeq 6000 Xp kits come in 2 configurations:

* **2 lane kits are for use with the S1 and S2 reagent kits.**
* **4 lane kits are for use with S4 kits.**

Each kit contains ExAmp reagents and a 2- or 4-lane manifold.

* **Note**: the reusable flow cell dock is sold separately.

* Xp flow cell dock is reusable.
* Flow cell is inverted and manifold is fitted over the flow cell.

**Xp Kit components, shipping conditions, and storage temperatures:**

| Quantity | Kit Component | Shipping Condition | Storage Temp |
| -------- | ------------- | ------------------ | ----------------------------------------------------------------- |
| 1 | DPX1/JPX1 | Dry ice | -20°C |
| 1 | DPX2/JPX2 | Dry ice | -20°C |
| 1 | DPX3 | Dry ice | -20°C |
| 1 | Xp Manifold | Dry ice | \*Can leave frozen with the kit or store at room temperature (RT) |

* Thaw components at RT for 10 min then set aside on ice.
* Mix and use within 30 minutes at room temperature or 1 hour if placed on ice.
* If unmixed with library pool, Xp reagents can be refrozen once (freeze as quickly as possible after thawing).
* Extra mastermix should be discarded.
* Wells on the manifold are numbered right to left to match the lane number of an inverted flow cell.
* Magnetic clamp holds in place when on dock.

**Workflow**

* Start the run within 30 minutes after loading library pools on the flow cell.
  * **Note**: prolonged incubation might reduce the percentage of clusters passing filter (%PF).
