What are the protocol differences when using single index and dual index adapters?
The amount of AMPure XP beads has been adjusted for the PCR clean-up. This has been optimized for the longer adapters (i.e. dual indexing adapters) present in the Adapter plate. What's the difference between the HS protocol and the LS protocol? The HS protocol requires additional peripheral equipment but reduces user touch-points and is ideally suited for projects that include >48 samples for the LT kit and >24 samples for the HT kit. The LS protocol requires minimal peripheral equipment but requires more user manipulation and is best suited for projects that include 48 or fewer samples for the LT kit and 24 or fewer samples for the HT kit. Can I use dual index adapters with the LS protocol? Can I use single index adapters with the HS protocol? Yes, the LS and HS workflows may be applied with both the single and dual index adapters. Following the recommended number of samples processed for each method depending on the kit used will ensure best results.
