# Library denaturation for the NextSeq 1000/2000

The NextSeq 1000/2000 system includes automated onboard denature and dilution of libraries, and an off-instrument, manual denaturation option.

**On-board denaturation**\
Denaturation and dilution occur within the reagent cartridge. Load double-stranded DNA when using the default on-board denature and dilute protocol.

**Off-instrument denaturation**\
If denaturing off-instrument, use Sodium hydroxide (NaOH) and time to complete the denaturation. For more information, search for Knowledge Base article **Manual Denaturation of Libraries on the NextSeq 1000/2000**.
