How to troubleshoot Insufficient Memory errors during Pre Run Checks on the NovaSeq X/X Plus

Background

Before starting a run, the Pre-Run Checks verify that the instrument is ready to perform a sequencing run and are split between the Dry and Wet checks. If the instrument does not have sufficient memory available to perform all of the necessary software actions during these Checks, it will present the following error:

Figure 1: Pre-Run Checks Errors - Insufficient memory to continue the execution of the program.

If the Pre-Run Checks are in the Dry Checks phase when the error occurs, the consumables will not be marked as used and are safe to be used in a follow up sequencing run.

If the Pre-Run Checks are in Wet Checks phase when the error occurs, the Flow Cell and Buffer Cartridge will be marked as used and new Flow cell and Buffer consumables will be required for a follow up sequencing run.

If this issue is observed, see the following steps to troubleshoot this issue.

Troubleshooting Actions

  1. Within the Control Software, select Return to Start to exit the Pre-Run Checks.

  2. Power cycle the instrument according to Illumina guidelines.

  3. Once the instrument has started and the Control Software is initialized, proceed with repeating the sequencing run setup.
