How to configure a NovaSeq 6000, with Control Software 1.8.x or greater, to work with a Proxy Server

After upgrading NovaSeq 6000 Control Software (NVCS) from version 1.7.5 or older, users may experience issues with utilizing the proxy server. Use this article to make the necessary configuration changes to continue utilizing the proxy server successfully. If the NovaSeq 6000 is configured to use a proxy server on NVCS 1.8+ without these changes, it is possible to experience the following error shortly after run start: "A component failed during sequencing and the run cannot be completed."

Change to use the IP address instead of Fully Qualified Domain Name (FQDN) in Manual Proxy Setup.

• In the Windows search bar, type Proxy Settings and select it.

  
• Scroll down to “Manual Proxy Settings” and confirm that the proxy’s IP address is provided, not the fully-qualified domain name (FQDN). If the FQDN is provided, please consult with your IT representatives to obtain the IP address and update this setting.

  

Change to use the IP address instead of FQDN in Internet Options

• In the Windows search bar, type Internet Options and select it.

  
• Select the Connections tab.

• Select LAN Settings.

  
• In the Proxy Server details section, confirm that the proxy’s IP address is provided, not the fully-qualified domain name (FQDN). If the FQDN is provided, consult the institutional IT representatives to obtain the IP address and update this setting.

  

Add the following environmental variables

• In the Windows search bar, type Environment Variables and select Edit the System Environment Variables.

  
• At the bottom of the window, select “Environment Variables”

  
• In the System Variables section, select New.

  • Input http_proxy as the variable name.

  • Input http://[IP Address of Proxy Server]:3128 as the variable value. - This should be the IP address of the Proxy server followed by :3128 (port). For example, http://12.34.56.78:3128

  
• In the System Variables section, select New.

  • Input https_proxy as the variable name.

  • Input http(s)://[IP Address of Proxy Server]:3128” as the variable value. - Work with local IT to determine if it should be http or https.

  
• In the System Variables section, select New.

  • Input no_proxy as the variable name.

  • Input localhost,127.0.0.0/8,169.254.0.0/16,127.0.0.1 as the variable value

  
• The following should now be listed in the System Variables section; select OK to apply the changes

  

Disable/re-enable Proactive in NVCS settings (if using Proactive)

• Disable Proactive.

  • Select the three lines in the top left of the control software, then Settings.

  • On the Mode Selection tab, at the bottom, uncheck the "Send Instrument Performance Data to Illumina" option, then select Save.

• Re-enable Proactive and select “save”

  • While still on the Mode Selection tab, at the bottom, check the "Send Instrument Performance Data to Illumina" option, then select Save.

Stop Illumina Universal Copy Service (UCS) in services.msc
