# Infinium HTS BeadChip Hybridization Loading Patterns for Manual and Automated Protocols

The following guidance outlines the Infinium High-Throughput Screening (HTS) BeadChip loading patterns for both the manual workflow and the automated workflow using the Tecan/Infinium Automated Pipetting System (IAPS) with Illumina Automation Control (IAC). Understanding the differences between these loading patterns is important for ensuring correct sample placement on BeadChips and assist with troubleshooting by determining whether observed failure patterns correlate with specific sample plate positions.# **Infinium HTS Manual Protocol**

**Single-Channel Pipette**

Using a single-channel pipette, transfer 14 µl of each sample from the MSA3 plate to the appropriate section of the BeadChip, as shown in Figure 1.

Ensure that the pipette tips are properly positioned within the sample inlets before dispensing the samples onto the BeadChip to prevent loading errors and ensure accurate sample distribution.

### **Multichannel Pipette**

Using an adjustable-spacing multichannel precision pipette, transfer 14 µl of each sample from the MSA3 plate to the appropriate section of the BeadChip, as shown in Figure 2.

Ensure that the pipette tips are properly positioned within the sample inlets before dispensing the samples onto the BeadChip to prevent loading errors and ensure accurate sample distribution.
