Kits, consumables, and reagent stability FAQ for Illumina Rapid RNA Kits

How many times can the Rapid RNA prep reagents be freeze/thawed?

Illumina has tested up to 6 freeze-thaw cycles with confirmed product performance.

What must be purchased for a Rapid RNA workflow? Are the kits bundled, or modular? If modular, what is needed?

  • The Rapid mRNA, Total RNA, and RNA with Enrichment are modular, and each Rapid RNA prep product page lists the components needed for the complete workflow.

  • The catalog products are bundled for the Rapid RNA Prep with Exome 2.5 Enrichment, VSPv2, RPIP, and RVEK, including the Rapid RNA prep, Fast Hyb Enrichment and Oligo panels.

  • Illumina DNA/RNA UD Indexes are sold separately for all workflows.

For Rapid Enrichment products, how many samples for library prep and how many enrichment reactions do the products support? Is this different from the non-Rapid versions?

Illumina Rapid RNA Prep with Enrichment(for custom enrichment panels):

  • Library prep reagents are sold for either 16 samples (20158823) or 96 samples (20158824)

  • Enrichment reagents are sold for either 16 samples (20040540, 16 hyb reactions), or for 96 samples (20040541, 32 hyb reactions)

  • Typically, users will purchase 20158823 and 20040540 to process 16 samples at 1-plex, or they will purchase 20158824 and 20040541 to process 96 samples at 3-plex.

Illumina Rapid Exome 2.5, VSPv2, RPIP, RVEK: These bundled kits support 96 samples, at 3-plex enrichments (32 enrichment reactions).

These plexities are the same as their non-Rapid versions, where applicable.

Compared to non-Rapid kits, are there any new consumables or equipment required for Rapid workflows?

Generally, compared to their non-Rapid versions, no new equipment or consumables are needed for the Rapid workflows. However, one notable difference is that users of the Illumina Rapid mRNA prep kit may need to use a microsample incubator to heat ST2 if there is precipitate. Additionally, Rapid RNA guides have been updated to have more comprehensive lists of required consumables (such as, gloves, lab tape, all pipette tip sizes, updated thermal cycler recommendations, etc.). Always consult the User-Supplied Consumables and Equipment section of the Product Documentation to verify all the required materials before starting library preparation.

Which indexes do Illumina Rapid RNA kits require? Where are their sequences listed?

Illumina DNA/RNA UD Indexes sets A-D, Tagmentation (cat # 20091654, 20091656, 20091658, 20091660, respectively). Index sequences can be found in the Adapter Sequences Document.

Do Rapid workflows utilize Unique Molecular Identifiers (UMIs)? If yes, where are they in the library?

No, Rapid workflows do not utilize UMIs.
