Troubleshooting Lyo Insert errors during run set up on the NovaSeq X Series

Errors when loading the Lyo Insert on the NovaSeq X/X Plus systems may display as:

  • "system could not read the lyo insert"

  • "unable to retrieve lyo insert information"

  • "the loaded lyo insert cannot support the loaded reagent cartridge"

This can result from control software version requirements, incompatible kit components, or consumable related issues.

Control Software Version Requirements

The 25B 100 cycle kit and 25B 200 cycle kit require control software version 1.3, refer to the v1.3 release notes. An error occurs when loading the Lyo Insert of 25B 100/200 cycle kit configurations on control software v1.2 and below. If a control software update is required, contact Illumina Technical Support.

Incompatible Kit Components

Confirm the Lyo Insert is from the correct reagent kit configuration. Lyo Inserts are not interchangable across 1.5B, 10B, and 25B kit configurations.

If an incorrect Lyo Insert is loaded on to the reagent cartridge and then loaded on the instrument, the control software recognizes the incorrect Lyo Insert and displays a warning that the loaded Lyo Insert does not match the proper cartridge configuration.

Note: The Lyo Insert from a 25B 100/200 cycle kit cannot be used with the 25B 300 cycle kits.

Consumable Related Issues

The affected Lyo Insert may have a consumable quality related issue. To troubleshoot:

  1. Re-load the Lyo Insert.

  2. If error persists after re-loading, use a new Lyo Insert.

  3. If the new Lyo Insert loads successfully, contact Illumina Technical Support with the failed consumable information (serial number, lot number, expiration date).
