# Recommended PhiX Loading for a NextSeq 550 Validation Run

A PhiX validation run confirms proper hardware and software performance of the instrument.

* Load 1.5pM PhiX for High-Output verification runs
* Load 1.5pM PhiX for Mid-Output verification runs

PhiX validation runs can be set up in manual mode or Local Run Manager

* For manual mode see [How to set up a PhiX validation run on the NextSeq 500/550 in manual mode](/instrumentation/nextseq-500-550/instrumentation-nextseq-500-550-reference_material-list/000001532.md)
* For Local Run Manager mode see [How to set up a PhiX validation run with Local Run Manager](/instrumentation/general/instrumentation-general-reference_material-list/000001318.md)

For loading concentrations on other platforms see [PhiX loading concentrations for validation runs on Illumina sequencing platforms](/instrumentation/general/instrumentation-general-reference_material-list/000001536.md)

# Agent Instructions: Querying This Documentation

If you need additional information that is not directly available in this page, you can query the documentation dynamically by asking a question.

Perform an HTTP GET request on the current page URL with the `ask` query parameter:

```
GET https://knowledge.illumina.com/instrumentation/nextseq-500-550/instrumentation-nextseq-500-550-reference_material-list/000002068.md?ask=<question>
```

The question should be specific, self-contained, and written in natural language.
The response will contain a direct answer to the question and relevant excerpts and sources from the documentation.

Use this mechanism when the answer is not explicitly present in the current page, you need clarification or additional context, or you want to retrieve related documentation sections.
