# Updating the DRAGEN Microbial Enrichment (DME) Databases on the MiSeq i100 Series

**Background**

The MiSeq i100 Series platform allows on-instrument DRAGEN analysis using a variety of DRAGEN Applications, including the DRAGEN Microbial Enrichment Plus (DME+) application. This Application requires specific databases are installed on the instrument through the Applications user interface, and must be installed manually after updating the DME+ Application.

See the following instructions for updating the DME+ Application and installing DME+ Database files on MiSeq i100 Series instruments.

**Resolution Actions**

*Pre-Update Activities*

1. Ensure that the **Illumina Software Update Manager (ISUM) v2.0** and **MiSeq i100 Series Digital Package v1.1** are installed.
   1. See instructions in the [MiSeq i100 Series Software v1.1.0 Release Notes & Update Instructions](https://support.illumina.com/downloads/miseq-i100-series-release-notes.html) for additional details if needed.
2. Navigate to the [MiSeq i100 Series Downloads](https://support.illumina.com/sequencing/sequencing_instruments/miseq-i100-plus/downloads.html) page, then download the following files under the **Workflow Installers** and **Microbial Database Installers** headers:
   1. DRAGEN Microbial Enrichment Plus (DME+) v1.1.7 Application .iapp file (Figure 1).
   2. DRAGEN Microbial Enrichment Plus (DME+) database .ires files as needed (Figure 2).
3. Transfer the **.iapp** and **.ires** files to an exFAT-formatted USB storage device or network-attached storage.

*Uninstall the Existing DME+ Database Files*

1. Login to the MiSeq i100 Series Control Software as a local **Administrator** user.
2. Select the **menu icon** in the top-left corner, then select **Settings**, then **Applications**.
3. Select the **DME+ v1.0.13 Application**.
4. On the **Configuration** page under the **Analysis** header, select the **trash can icon** to delete the Database.
5. Repeat **Step 4** for each currently installed Database.

*Uninstall the DME+ v1.0.13 Application*

1. Login to the MiSeq i100 Series Control Software as a local **Administrator** user.
2. Select the **menu icon** in the top-left corner, then select **Settings**, then **Applications**.
3. Select the **DME+ v1.0.13 Application**.
4. Select **Uninstall**, then follow the prompts to complete uninstallation of the **DME+ v1.0.13 Application**.

*Install the DME+ v1.1.7 Application*

Before performing the following steps, mount the exFAT-formatted USB storage device or network-attached storage under **External Storage**. See the [MiSeq i100 Series Documentation - External Storage](https://support-docs.illumina.com/IN/MiSeqi100Series/Content/IN/MiSeqi100/SysOutFolder.htm) for additional details.

1. Login to the MiSeq i100 Series Control Software as a local **Administrator** user.
2. Select the **menu icon** in the top-left corner, then select **Settings**, then **Applications**.
3. Select **Install**.
4. Navigate to the **DME+ v1.1.7 Application .iapp** file, then select **Open**.
5. The Control Software will then upload the file and display the **Application details** on completion.
6. Select **Install** and allow the Control Software to install the **DME+ v1.1.7 Application**.

*Install the DME+ Databases*

1. Login to the MiSeq i100 Series Control Software as a local **Administrator** user.
2. Select the **menu icon** in the top-left corner, then select **Settings**, then **Applications**.
3. Select the **DME+ v1.1.7 Application**.
4. On the **Configuration** page, select **Upload** file and then navigate to the **DME+ Database .ires file**, then select **Open**.
5. Follow the prompts to complete installation of the .ires Database file.
6. Repeat **Steps 4 and 5** for each Database file to be installed.
7. Select **Save**.

Once the Application update and associated Databases have been installed, **eject the USB storage device** or **remove the network-attached storage** under **External Storage** if no longer required.
