# Run Setup using the MiSeq i100 Series 100 cycle kit Turbo Custom Recipe

**Background**

The MiSeq i100 Series 100-cycle kit Turbo Custom Recipe supports an accelerated workflow for pathogen detection and identification, enabling 100 cycle single-end, dual-index runs to complete in 2.5-3 hours—compared to 4-5 hours for the standard MiSeq i100 Series recipes.

**Important Notes**

* The Turbo Custom Recipes are only compatible with **100-cycle kits**, though can be used with the 25M, 50M, or 100M flow cell configurations.
* The Turbo Custom Recipes have been tested for use with the [**Respiratory Pathogen ID/AMR Enrichment Panel (RPIP) Kit**](https://www.illumina.com/products/by-type/sequencing-kits/library-prep-kits/respiratory-pathogen-id-panel.html#tabgroup-1-tab2) (Illumina, Catalog 20047050)
* Users can use Turbo Custom Recipes with other Workflows or Applications, though Quality and Yield might be reduced and require optimization by the user.
* Run times with the Turbo Custom Recipes are optimized for **single**-**end**, **dual**-**index** configurations.
* Turbo Custom Recipe runs must be setup in Index First mode if index reads are performed.
* Paired-end runs are supported and will **add about 1 hour** to the total run time.
* The Turbo Custom Recipes require [**Control Software version 1.1.0**](https://support.illumina.com/sequencing/sequencing_instruments/miseq-i100-plus/downloads.html) or higher.
* The first few runs using the Turbo Custom Recipe will show projected run times consistent with Standard Recipe run times until the instrument completes several runs using the Turbo Custom Recipes.

**Run Setup Instructions**

1. Transfer the Turbo Custom Recipe files to a folder on a configured **External Storage** location, either network storage or USB drive, that is connected to the MiSeq i100 Series Instrument.

2. In the MiSeq i100 Series Control Software, select **Start**, then proceed with Run Setup using Run Planning, Sample Sheet, or Manual (BCL Only).

3. In the **Review run** user interface (UI), locate the Custom recipe section and click **Select**... (Figure 1).
4. In the **Select recipe file** UI, select the custom recipe file for the specific Flow Cell configuration used (Figure 2). Custom Recipe files are specific to the Flow Cell used, failure to select the correct Custom Recipe may result in Run failure.

* 25M/100cycle kit requires: 25M\_recipe-100-IF\_Turbo\_25M.xml
* 50M/100cycle kit requires: 50M\_recipe-100-IF\_Turbo\_50M.xml
* 100M/100cycle kit requires: 100M\_recipe-100-IF\_Turbo\_100M.xml

5. In the **Review run** UI, verify the Custom recipe is correct, then select **Load consumables** to proceed with Run Setup (Figure 3).
